Standard operating procedure for the detection of non-permitted rice varieties by indel-pcr analysis using the Agilent 2100 bionalyzer

Scope

The analysis uses a DNA-based method designed to detect permitted Basmati varieties and non-permitted varieties (non-Basmatis), in rice grain samples using PCR markers. It can be used to test uncooked or par-boiled samples of rice grains. It can identify whether a sample is a pure Basmati variety belonging to one of six groups, or whether it is a mixture or a non-permitted variety. It can detect the presence of non-permitted varieties in a mixture. The analysis comprises of two parts (i.e. two separate PCR reactions). The main method is suitable for the qualitative detection of seven possible adulterant rice varieties (not permitted for labelling as „Basmati‟). The optional follow-on tests can identify whether a sample is one of two further potential adulterants. All PCR products are analysed on the Bioanalyser.

Limitations: The test can not distinguish all Basmati varieties from each other. The main test can not detect the following non-permitted varieties: Yamini, Pusa 1121, Basmati 2000 and Shaheen Basmati. The follow –on tests can detect Pusa 1121 and Shaheen Basmati.
